There are five main subject areas in this volume in the series on medicinal chemistry. The first is a review of the understanding of Alzheimer's disease and the development of drugs for its treatment; the second, looking at recent efforts in modifying a naturally occuring anticancer (campothecin) for chemotherapy; the third covers the problem of getting a drug to a specific site within the context of phosphates and phosphonates; a survey of sterilization using aldehydes for the destruction of microbes both inside and outside the human body is reviewed in the fourth; and the last chapter is an account of the progress made in the biologically active enantiomer for complex synthetic asymmetric drug molecules.

Annotation The complex and multidisciplinary nature of modern drug discovery is well illustrated in the six chapters of this volume that describe exciting developments in both newly emerging and mature areas of medicinal chemistry. Chapter 1 provides a comprehensive review of therapeutic applications of ligands for peroxisome proliferator-activated receptor gamma PPAR?, a key regulator of glucose and lipid homeostasis. Progress over the last decade on ligands that bind to central nicotinic acetylcholine receptors is timely reviewed in Chapter 2, with the emphasis on the a4ß2 subtype. Chapter 3 reviews inhibitors of PARP-1, the founding member of the family, with the focus on their therapeutic potential in ischaemia related CNS injuries. Chapter 4 describes the structure-activity relationships of some semi-synthetic analogues including promising new compounds in development. Chapter 5 describes selective, first generation, non-peptidic neuropeptide Y1 and Y2 antagonists and their actions and evaluates their potential therapeutic application in cardiovascular disorders, with the emphasis on NPY1 antagonists. Chapter 6 is an extensive review of the considerable progress that has been achieved in developing Cathepsin K inhibitors and evaluates their potential for treating arthritis and atherosclerosis.

Volume 5 of Advances in Medicinal Chemistry contains four intriguing and detailed accounts of the close interface between synthetic chemistry, structure-activity relationships, biochemistry, and pharmacology. In Chapter 1, there is a comprehensive survey of the immunophilin area specifically focussing on neuroregenerative applications in the central nervous system. In Chapter 2, there is an overview of the development of a potent analgesic compound that works via modulation of neuronal nicotinic acetylcholine receptors. In Chapter 3, there is a description of dopamine D-2 autoreceptor partial agonists as potential therapy for the treatment of schizophrenia. In Chapter 4, there is a summary of the successful program in which potent non-peptide inhibitors of HIV protease from the AIDS virus were developed.

The modern pharmacopeia has enormous power to alleviate disease, and owes its existence almost entirely to the work of the pharmaceutical industry. This book provides an introduction to the way the industry goes about the discovery and development of new drugs. The first part gives a brief historical account from its origins in the mediaeval apothecaries’ trade, and discusses the changing understanding of what we mean by disease, and what therapy aims to achieve, as well as summarising case histories of the discovery and development of some important drugs. The second part focuses on the science and technology involved in the discovery process: the stages by which a promising new chemical entity is identified, from the starting point of a medical need and an idea for addressing it. A chapter on biopharmaceuticals, whose discovery and development tend to follow routes somewhat different from synthetic compounds, is included here, as well as accounts of patent issues that arise in the discovery phase, and a chapter on research management in this environment. The third section of the book deals with drug development: the work that has to be undertaken to turn the drug candidate that emerges from the discovery process into a product on the market. The definitive introduction to how a pharmaceutical company goes about its business of discovering and developing drugs. The second edition has a new editor: Professor Raymond Hill ● non-executive director of Addex Pharmaceuticals, Covagen and of Orexo AB ● Visiting Industrial Professor of Pharmacology in the University of Bristol ● Visiting Professor in the School of Medical and Health Sciences at the University of Surrey ● Visiting Professor in Physiology and Pharmacology at the University of Strathclyde ● President and Chair of the Council of the British Pharmacological Society ● member of the Nuffield Council on Bioethics and the Advisory Council on Misuse of Drugs. New to this edition: Completely rewritten chapter on The Role of Medicinal Chemistry in the Drug Discovery Process. New topic - DMPK Optimization Strategy in drug discovery. New chapter on Scaffolds: Small globular proteins as antibody substitutes. Totally updated chapters on Intellectual Property and Marketing 50 new illustrations in full colour Features Accessible, general guide to pharmaceutical research and development. Examines the interfaces between cost and social benefit, quality control and mass production, regulatory bodies, patent management, and all interdisciplinary intersections essential to effective drug development. Written by a strong team of scientists with long experience in the pharmaceutical industry. Solid overview of all the steps from lab bench to market in an easy-to-understand way which will be accessible to non-specialists. From customer reviews of the previous edition: ‘... it will have everything you need to know on this module. Deeply referenced and, thus, deeply reliable. Highly Commended in the medicine category of the BMA 2006 medical book competition Winner of the Royal Society of Medicine Library Prize for Medical Book of the Year

The success of any drug discovery project relies upon the quality of the lead that initiates the lead optimization process. What defines a ‘quality lead’, where these ‘quality leads’ come from and how one discovers them has been the subject of intense debate within the pharmaceutical industry, relies upon defining those properties that historically have led to successful drug discovery. This volume addresses these questions and specifically discusses diabetes, obesity and tuberculosis. *Presents the latest research in the field of drug discovery *Publishes on an annual basis to bring you the most innovative updates in medicinal chemistry *Available as an online resource via ScienceDirect
